Persicaline, an alkaloid from <i>Salvadora persica</i>, inhibits proliferation and induces apoptosis and cell-cycle arrest in MCF-7 cells

Abstract Cancer is the second largest cause of mortality worldwide. Many natural bioactive chemicals generated from plants have favorable impacts on health, including cancer chemoprevention, compared to their manufactured counterparts. Persicaline, a novel sulfur-containing imidazoline alkaloid derived Salvadora persica , has been shown display promising antioxidant activity. In this study, antiproliferative activity persicaline was tested against different cells using (3-[4,5-dimethylthiazol-2-yl]-2,5 diphenyl tetrazolium bromide assay. The cell death mode and cell-cycle arrest were examined flow cytometry analysis. addition, proapoptotic molecular mechanism effects mammary MCF-7 line explored. Furthermore, impact apoptotic genes markers, generation reactive oxygen species (ROS), mitochondrial membrane potential monitored. It found that inhibits proliferation in dose-dependent manner. Persicaline-treated also showed initiation events G1 arrest. treatment led an increase ROS generation, Bax caspase upregulation while Bcl-2 downregulated. Hence, for first time, study causes phase apoptosis induction cells. Increased levels required persicaline.
